Support for SermonSpeaker 5.x is in bugfix mode only, the same as it is for Joomla 3.10.
I will not add any new features to this releases.
Please upgrade to SermonSpeaker 6.x and Joomla 4.x, which is better anyway.

I'm uploading my weekly sermon as normal (to Amazon server), but although I can see the mp3 file uploading with the progress bar, when it gets to the end, it does nothing - i.e. doesn't appear in the field above. The file is only 30mb so i don't see that as the issue. Do you think this is a SermonSpeaker issue or do you think there could be a problem with the Amazon Servers?

If you don't get an error at all, I would suspect a server issue. The apache error log would likely tell you more.
At least if you didn't change anything lately and it worked the week before.

Thanks Thomas, tried it again - still didn't work, returned to the screen after a while and the following error appears now in a pink box:

Error #-200: HTTP Error.

Does that point us in any direction to rectify the situation?

Not really. The code would actually say it was a success (HTTP Code 200 is "OK").
Can you check the apache error log? You can also try to enable error reporting within Joomla, but you still need to check the error log.
If you don't have access to it, you will have to ask your hoster.
If you didn't change anything in your setup and were able to upload files of that size before, then you will have to ask your hoster anyway. It's likely that they changed a setting somehwere.

Thanks again Thomas. For your information, I got the following response from my host (the file appears to upload as normal now, but I have to wait about a minute for the file name to actually show in the field, but at least it works again)...

If you would like to see any errors which have been recorded in the Apache server logs, you can view them from your cPanel --> Error Log.

I have noticed that there is a timeout on the following script:

It seems that the file you are trying to upload is reaching the server's timeout. I have dynamically increased the timeout for this particular script. I would suggest you to try uploading the file once again. If this workaround does not help, I would suggest you to upload the file in smaller bulks or use an FTP client in order to upload the file to the desired folder.

Please note that the timeout can not be changed due to the fact that your account is hosted in a shared environment. This setting can only be modified on our Cloud/Dedicated servers.

When it hits a timeout, it's likely a connection issue. Like you're upload bandwith is limited somewhere.

The delay between the upload finishing in the form and the filename to appear may be caused by the further processing of the file. In your case it's first uploaded to your server and from there it will be uploaded to Amazon S3. So basically it's uploaded twice :)
