Support for SermonSpeaker 3.x is canceled as it is for Joomla 1.5. I will gladly assist you with minor issues but I will not fix any bugs anymore in this releases.
The fix is to upgrade to SermonSpeaker 4.x, which is better anyway.

Having issue with uploading sermons was created by Marvin Serwanga
This is the filepath: /images/sermons/Pastor_Andrew_The_Key_Of_Thanksgiving.mp3 but when I click on the published sermon I get the 404 error.

Looks like there are spaces in the filename in your picture.
You can also try lowercasing the whole filename.

And of course update your Joomla to the latest version (3.6.5). There were several critical security issues which are not fixed in 2.5 since that one is end of life.

OK I tried lower case and removing spaces still having issues with uploading the sermon to the site.

Then update Joomla and SermonSpeaker. Sorry to say but if you're still using SermonSpeaker 3.x (like indicated by this forum choice) then you have much bigger troubles than that single file that doesn't play. You're running a piece of software that is at least 4 years old and likely vulnerable to security issues.
Chances are actually high that your site is already hacked.
